Contraindications

Warnings and Precautions
• Severe allergies: Permanently disable LuciAlpe. Start appropriate treatment immediately.
• severe skin adverse reactions (scar): LuciAlpe can cause scar, including Stevens-Johnson syndrome (SJS), erythema multiforme (EM), toxic epidermal necrolysis (ten), and drug reactions with eosinophilia and systemic symptoms (dress). If you have signs or symptoms of scar, interrupt LuciAlpe. If scar is confirmed, permanently deactivate LuciAlpe.
• Hyperglycemia: LuciAlpe can cause severe hyperglycemia, which in some cases is associated with hyperglycemic hyperosmolar non-ketotic syndrome (HHNKS) or ketoacidosis. The safety profile of LuciAlpe in patients with type 1 or uncontrolled type 2 diabetes has not been determined. Test fasting plasma glucose (FPG), HbA1c, and optimize blood glucose before starting treatment with LuciAlpe. After starting treatment, monitor regularly. Start or optimize antihyperglycemic medications as clinically indicated. In case of severe hyperglycemia, interrupt, reduce the dose or discontinue LuciAlpe.
• Pneumonia: LuciAlpe can cause severe pneumonia and interstitial lung disease. Monitor for clinical symptoms or radiological changes. In case of severe pneumonia, interrupt or discontinue LuciAlpe.
• Diarrhea or colitis: LuciAlpe causes diarrhea in most patients and can be severe, leading to dehydration and acute kidney damage. Patients are advised to start diarrhea treatment, increase oral fluids, and notify their healthcare provider in case of diarrhea.
Colitis has been reported in patients treated with LuciAlpe. Monitor for other symptoms of diarrhea and colitis, including abdominal pain and mucous or bloody stools.
In case of severe diarrhea or colitis, interrupt, reduce dose or discontinue LuciAlpe. Patients with colitis may require additional treatment, such as enteric-coated steroids and/or systemic steroids.
• Embryo-fetal toxicity: LuciAlpe can cause fetal harm. Patients are advised to be aware of potential risks to the fetus and to use effective contraception.

Adverse Reactions
The most common adverse reactions, including laboratory abnormalities (all grades, incidence ≥ 20%) are increased glucose, increased creatinine, diarrhea, rash, decreased lymphocyte count, increased gamma-glutamyl transferase (GGT), nausea, increased alanine aminotransferase (ALT), fatigue, decreased hemoglobin, increased lipase, decreased appetite, stomatitis, vomiting, decreased weight, decreased calcium, decreased glucose, prolonged activated partial thromboplastin time (aPTT) Long and hair loss.
