Contraindications

Warnings and Precautions
• Diarrhea: Prevention and control of diarrhea by increasing the dose of LuciNera or loperamide. If diarrhea occurs despite recommended precautions, use additional antidiarrheals, fluids, and electrolytes as clinically indicated. LuciNera should be discontinued for patients with severe and/or persistent diarrhea. LuciNera should be permanently discontinued for patients with grade 4 diarrhea or grade ≥ 2 diarrhea after maximum dose reduction.
• Hepatotoxicity: Hepatic function is monitored monthly for the first 3 months of treatment and then every 3 months during treatment and as clinically indicated. LuciNera should be interrupted in patients with grade 3 liver abnormalities and permanently discontinued in patients with grade 4 liver abnormalities.
• Embryo-fetal toxicity: LuciNera can cause fetal harm. Patients are advised to be aware of potential risks to the fetus and to use effective contraception.

Adverse Reactions
The most frequent adverse reactions (reported in ≥ 5% of patients) were:
• When LuciNera is used alone: diarrhea, nausea, abdominal pain, fatigue, vomiting, rash, stomatitis, decreased appetite, muscle spasms, dyspepsia, increased AST or ALT, nail disease, dry skin, abdominal distension, nasal bleeding, weight loss, and urinary tract infections.
• When LuciNera is used in combination with capecitabine: diarrhea, nausea, vomiting, decreased appetite, constipation, fatigue/weakness, weight loss, dizziness, back pain, joint pain, urinary tract infection, upper respiratory tract infection, bloating, renal impairment, and muscle spasms.
