Contraindications

Warnings and Precautions
• Hepatic adverse reactions were reported with the combined regimen of Pretomanid tablets, bedaquiline and linezolid. Monitor for signs and symptoms and laboratory tests related to the liver. If there are signs of liver damage, interrupt the entire regimen.
• Combination regimens with Pretomanid tablets, bedaquiline, and linezolid have been reported to inhibit bone marrow hyperplasia. Monitor the complete blood count. Reduce or interrupt linezolid dosing in case of severe or worsening myelosuppression.
• Peripheral nerve and optic neuropathy was reported with the combination regimen of Pretomanid tablets, bedaquiline and linezolid. Monitor visual functions. If there are symptoms of visual impairment, an ophthalmological assessment should be performed. Reduce or interrupt linezolid dosing if neuropathy develops or worsens.
• Prolonged QT interval with combination regimen of Pretomanid tablets, bedaquiline and linezolid. Use with medications that prolong the QT interval may result in prolongation of the QT interval. Monitor the ECG. In case of severe ventricular arrhythmias or prolongation of the QTcF interval by more than 500 msec, the combination regimen of Pretomanid tablets, bedaquiline and linezolid will be discontinued.
• Reproductive effects: Pretomanid causes testicular atrophy and impaired fertility in male rats. Inform patients that the reproductive toxicity found in animal studies, as well as the potential effects on male fertility in humans, has not been fully evaluated.
• Lactic acidosis has been reported with the combined regimen of Pretomanid tablets, bedaquiline and linezolid. In case of severe lactic acidosis, consider interrupting the entire combination regimen with linezolid or Pretomanid tablets, bedaquiline and linezolid.

Adverse Reactions
The most common adverse reactions (≥ 10%) are peripheral neuropathy, acne, anemia, nausea, vomiting, headache, elevated aminotransferase, dyspepsia, decreased appetite, rash, itching, abdominal pain, pleurisy pain, elevated gamma-glutamyltransferase, lower respiratory tract infection, hyperamylasemia, hemoptysis, back pain, cough, impaired vision, hypoglycemia, abnormal weight loss, and diarrhea.
