Contraindications

Warnings and Precautions
• Concurrent use of a potent CYP3A inducer carries a serious risk of hepatotoxicity: prior to starting luciLorla, the potent CYP3A inducer should be discontinued within 3 plasma half-lives of the potent CYP3A inducer.
• Central nervous system (CNS) effects: CNS effects include seizures, psychotic effects, and changes in cognitive function, mood (including suicidal ideation), speech, mental state, and sleep. Suspend and resume the same or reduced dose of luciLorla, or permanently discontinue luciLorla, depending on the severity.
• Hyperlipidemia: Start or increase the dose of lipid-lowering agents. Suspend and resume the same or reduced dose of luciLorla, depending on the severity.
• AV block: pause and resume the same or reduced dose of luciLorla, depending on the severity.
• Interstitial lung disease/pneumonia: Immediately discontinue use of luciLorla in patients with suspected interstitial lung disease/pneumonia. For treatment-related interstitial lung disease/pneumonia of any severity, permanently discontinue luciLorla.
• Blood Pressure: Blood pressure is monitored after 2 weeks and then at least monthly during treatment. For severe blood pressure, discontinue luciLorla and then reduce the dose or permanently discontinue.
• Hyperglycemia: Fasting blood glucose is regularly assessed before and during treatment with luciLorla. If not adequately controlled by optimal medical management, discontinue use of luciLorla and then consider dose reduction or permanent discontinuation, depending on the severity.
• Embryo-fetal toxicity: can cause fetal harm. Women of childbearing potential are advised to be aware of the potential risks to the fetus. Effective non-hormonal methods of contraception are recommended for men and women of childbearing potential.

Adverse Reactions
The most common (incidence ≥ 20%) adverse reactions and grade 3-4 laboratory abnormalities are edema, peripheral neuropathy, weight gain, cognitive effects, fatigue, dyspnea, arthralgia, diarrhea, emotional effects, hypercholesterolemia, hypertriglyceridemia, and cough.
