Contraindications

Warnings and Precautions
• Thrombocytopenia: Platelet counts are monitored throughout treatment. Managed through discontinuation and/or dose reduction and supportive care.
• Neutropenia: Neutrophil counts are monitored throughout treatment. Management by discontinuation and/or reduction of dose and granulocyte colony-stimulating factor.
• Gastrointestinal toxicity: Nausea, vomiting, diarrhea, anorexia, and weight loss may occur. Provides antiemetic prophylaxis. Managed through dose interruption and/or reduction, antiemetics, and supportive care.
• Hyponatremia: Serum sodium levels are monitored throughout treatment. Correcting concomitant hyperglycemia and high serum paraprotein levels. Managed through interruptions, reductions or discontinuations, and supportive care.
• Serious infections: Monitor for infections and treat them promptly.
• Neurotoxicity: Patients are advised not to drive and engage in hazardous occupations or activities until neurotoxicity has resolved. Optimize hydration status and concomitant medication to avoid dizziness or mental state changes.
• Embryo-fetal toxicity: May cause harm to the fetus. Men of women of childbearing potential and of female partners of childbearing potential are advised to be aware of the potential risks to the foetus and to use effective contraception.
• Cataracts: Cataracts may develop or progress. Treatment of cataracts often requires surgical removal of the cataract.

Medication risks

Adverse Reactions
• The most common adverse effects (≥ 20%) in patients with multiple myeloma treated with XVd are fatigue, nausea, decreased appetite, diarrhea, peripheral neuropathy, upper respiratory tract infection, weight loss, cataracts, and vomiting. Grade 3-4 laboratory abnormalities (≥ 10%) are thrombocytopenia, lymphopenia, hypophosphatemia, anemia, hyponatremia, and neutropenia.
• The most common adverse effects (≥ 20%) in patients with multiple myeloma treated with Xd are thrombocytopenia, fatigue, nausea, anemia, decreased appetite, weight loss, diarrhea, vomiting, hyponatremia, neutropenia, leukopenia, constipation, dyspnea, and upper respiratory tract infections.
• In addition to laboratory abnormalities, the most common adverse reactions (incidence ≥ 20%) in patients with DLBCL are fatigue, nausea, diarrhea, decreased appetite, weight loss, constipation, vomiting, and fever. Grade 3-4 laboratory abnormalities (≥ 15%) are thrombocytopenia, lymphopenia, neutropenia, anemia, and hyponatremia.
