Contraindications

Warnings and Precautions
• Central nervous system (CNS) effects: can cause adverse CNS reactions, including dizziness, ataxia, and cognitive impairment. Discontinue the medication and resume at the same or reduced dose after your condition improves, or permanently discontinue LuciRepo depending on the severity.
• Interstitial lung disease (ILD)/pneumonia: Monitor patients for new or worsening lung symptoms that indicate ILD/pneumonia. Immediately discontinue treatment for patients with suspected ILD/pneumonia or permanently if ILD/pneumonia is confirmed.
• Hepatotoxicity: Hepatic function is monitored every 2 weeks during the first month of treatment and thereafter as clinically indicated. Depending on the severity, interrupt, then resume with the same or reduced dose, or permanently discontinue.
• Myalgia with elevated creatine phosphokinase (CPK): Monitor serum CPK levels in patients who report unexplained muscle pain, tenderness, or weakness during treatment. Depending on the severity, discontinue and resume at the same or reduced dose once the condition has improved.
• Hyperuricemia: Serum uric acid levels are regularly monitored before and during treatment. Start treatment with uric acid-lowering medications as clinically indicated. Depending on the severity, discontinue and resume with the same or reduced dose, or permanently discontinue.
• Fractures: Patients with signs or symptoms of fractures (e.g., pain, changes in activity, deformities) are assessed in a timely manner.
• Embryo-fetal toxicity: can cause fetal harm. Women of childbearing potential are advised to be aware of the potential risks to the fetus and to use effective non-hormonal methods of contraception.

The most common adverse reactions (≥ 20%) are dizziness, taste disorders, peripheral neuropathy, constipation, dyspnea, ataxia, fatigue, cognitive impairment, and muscle weakness.
