Contraindications

Warnings and Precautions
• Blood Pressure: Monitor blood pressure and control blood pressure as clinically indicated. If Blood Pressure cannot be medically controlled, interrupt, reduce dose, or discontinue LuciPona.
• Pancreatitis: Monitor for serum lipase. Interrupt, then resume with the same or reduced dose, or discontinue LuciPona depending on the severity. Pancreatitis is assessed when elevated lipase is accompanied by abdominal symptoms.
• Neuropathy: Monitor for symptoms of peripheral neuropathy and cerebral neuropathy. Interrupt and then resume at the same or reduced dose or discontinue LuciPona depending on recurrence/severity.
• Eye Toxicity: Comprehensive eye examinations are performed regularly at the beginning and during treatment.
• Bleeding: Monitor bleeding and manage patients as clinically indicated. Interrupt and then resume at the same or reduced dose or discontinue LuciPona depending on recurrence/severity.
• Fluid retention: Monitor fluid retention and manage patients as clinically indicated. Depending on the recurrence/severity, interrupt and then resume or discontinue LuciPona at the same or reduced dose.
• Arrhythmia: Monitor for signs or symptoms of arrhythmia and manage patients as clinically indicated. Depending on the recurrence/severity, interrupt and then resume or discontinue LuciPona at the same or reduced dose.
• Myelosuppression: CBC every 2 weeks for the first 3 months, then monthly or as clinically indicated. If ANC is less than 1 x 109/L or platelets are less than 50 x 109/L, interrupt LuciPona until ANC reaches at least 1.5 x 109/L and platelets at least 75 x 109/L, then resume at the same or reduced dose.
• Tumor lysis syndrome: Ensure adequate hydration and correct elevated uric acid levels before starting LuciPona.
• Reversible posterior leukoencephalopathy syndrome (RPLS): interrupt LuciPona until cured. The safety of returning patients to LuciPona after recovery from RPLS is unknown.
• Impaired wound healing and gastrointestinal perforation: discontinue use of LuciPona at least 1 week prior to elective surgery. Do not use LuciPona for at least 2 weeks after major surgery and until the wound has fully healed. The safety of returning to LuciPona after healing from wound healing complications has not been determined.
• Embryo-fetal toxicity: May cause harm to the fetus. Women of childbearing potential are advised to be aware of the potential risks to the fetus and to use effective contraception.

Medication risks

Adverse Reactions
The most common (> 20%) adverse reactions are rash and related symptoms, arthralgia, abdominal pain, headache, constipation, dry skin, Blood Pressure, fatigue, fluid retention and edema, pyrexia, nausea, pancreatitis/lipase increase, bleeding, anemia, liver dysfunction, and AOEs. The most common grade 3 or 4 laboratory abnormalities (> 20%) are platelet count decreased, neutrophil count decreased, and leukopenia.
