Contraindications

CAUTION:
Adrenal crisis: Patients using MITOSIS may experience an adrenal crisis in the event of shock or severe trauma, and the response to shock may be diminished. Hydrocortisone should be given, signs of increased shock should be monitored, and treatment with MITOSIS should be interrupted until the patient recovers.

Central Nervous System Toxicity: Treatment with MITOSIS may cause central nervous system toxicity, including drowsiness, lethargy, and dizziness. When the plasma concentration of mitotane exceeds 20 mcg/mL, the incidence of toxicity is higher.

Adrenal insufficiency: Treatment with MITOSIS may result in adrenal insufficiency. Hormone replacement therapy is performed as clinically necessary and optimized by measuring free cortisol and adrenocorticotropic hormone (ACTH) levels.

Pregnant woman medication warning: MITOSIS may cause harm to the fetus. Patients exposed to mitotane during pregnancy may experience abnormal pregnancy outcomes, such as premature birth or early miscarriage. Pregnant women are advised to be aware of the potential risks to the fetus, and women of childbearing potential are advised to use effective contraception during treatment and for as long as mitotane plasma levels are detectable after discontinuation.

Ovarian cysts: Bilateral and multiple ovarian cysts may occur in women of childbearing age treated with MITOSIS. These cysts can lead to complications such as attachment torsion and bleeding from ruptured cysts. In some cases, improvement is seen after discontinuation of MITOSIS. Female patients are advised to seek medical attention in time for gynecological symptoms such as vaginal bleeding and/or pelvic pain.
